PDF layouts for PCB's

I read the posts on toner transfer pcb printing on this forum and would like to try my hand at this. When I open the pages to look at the PCB it usually show 150% , 101% etc. I have two questions on this.

Should I select 100% to get the correct size of layout to work with and should these be printed in reverse?

select 100% when printing. Any box that lets you resize to fit, turn off. Toner transfer requires a mirror image which is what the pdf's are.
